- W2081120192 abstract "Abstract A temperature-sensitive mutant of Semliki Forest virus has been shown to induce the synthesis of two short-lived nonstructural proteins with molecular weights approx. 155 000 (ns155) and 135 000 (ns135), and four smaller proteins with molecular weights 86 000 (ns86), 72 000 (ns72), 70 000 (ns70) and 60 000 (ns60). Tryptic peptide mapping of the [35S]methionine-labelled proteins revealed that certain reproducible peptides in ns155 were also regularly detected in ns86. Other peptides in ns155 could be detected in cytoplasmic extracts pulse-labelled for 3 min after release of protein synthesis from the hypertonic initiation block. These peptides were also detected in material migrating with a molecular weight of 70 000–72 000, and are therefore considered to be specific for ns70. The peptide map derived from ns135 was different from that derived from ns155: a characteristic group of peptides in ns135 was also detected in the mixture of proteins with molecular weight 70 000–72 000 and so must be derived from ns72. It was not possible to identify the peptides specific for ns60. The results confirm the precursor-product relationship proposed earlier (Lachmi, B. and Kaariainen, L. (1976) Proc. Natl. Acad. Sci. U.S. 73, 1936–1940). A scheme for the translation and cleavage of the non-structural proteins of Semliki Forest virus is presented." @default.
