- W2081839178 abstract "Patients suffering from heat stroke usually present a systemic inflammatory response possibly caused by the transposition of bacterial lipopolysaccharide (LPS) from the gut to the systemic circulation. It has been shown that increases in intracellular heat shock protein 72 (Hsp72) reduces the release of pro-inflammatory cytokines in macrophages stimulated with LPS. In humans, whole body heat acclimation (HA) induces increases in Hsp72 in peripheral blood mononuclear cells (PBMC). However, it is not known if HA can modify the release of cytokines in PBMC stimulated with LPS. PURPOSE: To determine if HA modifies the PBMC release of cytokines when stimulated with LPS in vitro. We hypothesized that HA would reduce the PBMC release of pro-inflammatory cytokines (TNF-α, IL-1 and IL-6) and increase anti-inflammatory cytokine (IL-10) when stimulated with LPS. METHODS: 7 male and 2 female subjects (mean ± SD, age 24 ± 3 yrs, body weight 75 ± 5 kg) were heat acclimated by walking/running (56% VO2max) for 100 minutes in a hot environment (42°C) for 10 days. Blood was drawn, and PBMC were separated and cultured on days 1 and 10 of HA. 24 hours later, PBMC were divided into two groups: control (CON) or LPS (1 ng.mL-1 / 24 hours). Cytokine release into the cell media and Hsp72 were assayed by ELISA. RESULTS: As expected, HA induced a reduction in heart rate and core temperature. PBMC Hsp72 was higher (p<0.05) on day 10 than day 1 of HA. LPS exposure induced a greater cytokine release than CON but HA did not alter this response (table 1)Table 1: Cytokines concentrations in pg/mL. *indicate difference between CON and LPSCONCLUSION: HA increases Hsp72 but it does not modify the in vitro cytokine release to LPS.
