- W2083318954 abstract "There have been considerable problems in relating changes in behavior to the extent of neuronal degeneration after lesions of central monoamine pathways. Dopamine (DA) denervated animals are almost inaccessible to behavioral analysis because of their serious akinesia. We have developed an underwater swim test where the DA denervated animals are able to overcome this akinesia. Their swim speed is greatly reduced compared to normal animals and we found swim speed to be correlated to the extent of DA loss. Rats were denervated by intracerebral 6-hydroxydopamine injections, tested for their swim speed 48 h later when the terminal degeneration was complete and kept alive by tube feeding for a total of 8–9 days to allow for retrograde degeneration of DA cell bodies. DA levels were measured biochemically in caudate nucleus and the number of remaining DA cells were estimated in the A9 and A10 nuclei by counting fluorescent DA cells in sections 100 μm apart. Highly significant correlations were found between log number of DA cells in A9 (substantia nigra) and swim time, log % DA remaining in the caudate nucleus and swim time, and finally between number of remaining A9 cells and % DA remaining in the caudate nucleus. No such correlation existed with the DA cells in the A10 cell group. The method of counting cells seems to be a highly sensitive way to determine the extent of denervation in the nigrostriatal DA system and the swim speed test gives a sensitive indication of the extent of denervation below 10 % of normal values.
