We have particularly described the methodologies suitable for the purification of nucleic acids (DNA and RNA) and proteins interacting with and involved in the mechanisms of transcription and translation (enzymes, factors, . . . ). This work, the aim of which is the elucidation of the molecular mechanisms by the isolation of some key factors, has since then been widely expanded in many laboratories, as shown by recent publication@. In connection with this, affinity chromatography has only had an analytical or micropreparative role, which will be detailed below. Moreover, molecular genetics is a discipline upstream of cellular biology, implying that it allows modification of the expression mechanism in order to direct protein biosynthesis according to specific needs. Indeed, cellular biology is already integrated into biotechnology of high-value-added proteins, such as in preparative and industrial aims. It is only a means of attaining the chosen objective (production of a given protein), but cellular biology itself and, particularly, cell culture represent major axis of research and development ." @default.
