A mixture of this cholesterol and [4- 14 C]cholesterol was fed to guinea-pigs for 3 weeks. 2. 2. The cortisol and 6β-hydroxycortisol isolated from the urine of these animals were labeled with 14 C but not with 3 H. 3. 3. When an acetone-washed, beef adrenal gland mitochondrial fraction was incubated with [4- 14 C, 4β- 3 H]cholesterol, pregnenolone in which 3 H was retained and progesterone practically devoid of 3 H were isolated from the incubation mixture. 4. 4. The loss of the 4β-hydrogen in experiments both in vivo and in vitro is attributed to adrenal gland Δ 5 -3-ketosteroid isomerase (3-ketosteroid Δ 4 - Δ 5 -isomerase, EC 5.3.3.1) action. 5. 5. Two tentative schemes for adrenal gland Δ 5 -3-ketosteroid isomerase action are advanced, either of which could account for loss of the 4β-hydrogen." @default.
