- W2085702562 abstract "Abstract Pyrethroid-induced motor symptoms, i.e. deltamethrin-induced writhing and cismethrin-induced tremor, were studied using a number of pharmacological agents, in intact conscious rats and spinal rats. In intact conscious rats, procainamide, l - and d -propranolol and mephenesin modified both types of motor symptoms, while baclofen, meprobamate and phenytoin were ineffective. Mephenesin prevented or reversed all motor symptoms, whereas propranolol and procainamide were only effective in reducing the severity of motor symptoms, when given prior to the pyrethroids. In the spinal rat, intravenous injection of deltamethrin or cismethrin, elicited writhing or tremor, respectively. When using doses of mephenesin or propranolol in the spinal rat which were similar to those used in the intact rat, equivalent modification of motor symptoms were observed. Mephenesin was also used in investigations of some “non-motor” symptoms produced by deltamethrin, namely increased brain blood flow in the intact rat, and rise in blood glucose in the intact and spinal rat, and cardiovascular changes in the spinal rat. In the intact rat, doses of mephenesin which inhibited writhing did not modify the increases in blood flow in the brain or blood glucose following injection of deltamethrin. In the spinal rat mephenesin, at doses which prevented writhing, did inhibit the rise in blood glucose and the cardiovascular changes which follow deltamethrin. In conclusion it is suggested that pyrethroid-induced motor symptoms, i.e. writhing and tremor, are mediated via a spinal site of action probably involving interneurones. Deltamethrin-induced “non-motor” symptoms, i.e. increase in brain blood flow and blood glucose may result from a supra spinal component of deltamethrin activity. In contrast the cardiovascular effects of deltamethrin are mediated via a peripheral site of action." @default.
