- W2086211019 abstract "Culture conditions ofPyricularia oryzae Cav. protoplasts were examined to obtain a maximum level of reversion to hyphae, and a thin-layer agar-plating method was devised for the quantitative evaluation of events associated with protoplast regeneration and reversion. Reversion was categorized into four types based on abnormal budding and formation of hyphae from either budded cells or mother cells with regenerated cell walls. Budding and reversion to hyphae from budded cells were prevalent in liquid media, while the direct emergence of hyphae from mother cells with regenerated cell walls occurred most frequently in hard media containing 2% agar. The reversion types were mainly dependent on the agar concentration of the medium: the direct emergence of hyphae from mother cells with regenerated cell walls increased in frequency with the agar concentration, while the frequency of budding was negatively correlated with the agar concentration. The thin-layer agar-plating method gave a reversion frequency of 20–24% on a number of media tested, including potato-glucose, Vogel's, and Czapek. Cell wall regeneration and reversion were more quickly achieved in thin-layer agar-plate culture than in liquid culture. The use of the thin-layer agar plates with either potato-glucose or Vogel's medium is useful for numerical evaluation of regeneration and reversion ofP. oryzae protoplasts." @default.
