- W2087180568 abstract "The effect of selective removal of histone proteins from chicken erythrocyte nuclei on the process of reversible dispersal of chromatin in low ionic strength was examined in electron and light microscopes. Unextracted nuclei showed complete dispersion of chromatin in water and re-establishment of condensed structure on return to saline. After selective extraction of histones I and V at pH 1.8 and complete extraction of all histones at pH 1.0, nuclei held in saline still showed remnants of condensed chromatin structure which dispersed in water and reformed on return to saline. In the light microscope, both unextracted and extracted nuclei, when returned to saline after water exposure, showed precise re-establishment of the patterns of chromatin condensation present prior to water treatment. It is suggested that histone proteins play only a secondary role in gross chromatin organization, with nuclear membrane, non-histone proteins and DNA sequence being more centrally important. Histones are presumed to be of prime importance in the fine organization of individual chromatin fibrils. Chromatin in isolated liver nuclei also showed reversible dispersal in response to ionic strength although pattern re-establishment was less precise than in erythrocyte nuclei. A general model of chromosomal organization in interphase is proposed wherein individual chromsomes, each consisting of a single nucleoprotein fibril, are anchored to the nuclear membrane at numerous points." @default.
