We have studied the DNA reassociation kinetics in 6 new reptile species and, in addition, we have carried out a comparative analysis of the genome composition of 7 turtles and 7 scaly-reptiles. 2. In scaly-reptiles, as the phyletic distance increases, the differences in the most heterogeneous fractions (middle repetitive and single-copy DNAs) increase, too. In turtles such an increase is not found, but remarkable differences in single-copy DNA are observed also in congeneric species. 3. Moreover, in Squamata as the genome size increases, repetitive DNA increases more drastically than single-copy DNA, whereas in Chelionia both fractions increase with the same rapidity. 4. It is suggested that in turtles DNA evolution has followed a trend where deletion events have been very rare. In squamates, instead, there would have been a greater balance among amplifying events, divergences in the repetitive sequences and deletion. 5. These results are in agreement with the hypotheses that turtles and squamates belong to two clearly distinct branches of amniotes." @default.
