- W2088133602 abstract "Abstract Recent investigations have indicated the presence of a high-affinity binding site in brain for the benzodiazepines. GABAer-gic agonists enhance and antagonists inhibit binding to this high-affinity site, and binding can be correlated with effects on neuronal cell firing in the dorsal raphe nucleus. Certain anions (such as chloride, iodide, etc.) also enhance (3H)-diazepam binding, and this enhancement is consistent with their role in presynaptic inhibition. Taken together, these results suggest the existence of a GABA/C1- ionophore/BZ binding complex in brain. Attempts are underway to purify some of the components of this complex. By use of detergent (Lubrol) extraction, a soluble preparation of benzodiazepine receptors has been obtained. It has been suitable for receptor purification, and a radioactive derivative is in preparation. After solubilization, the receptor possesses the same pharmacological profile as the membrane-bound form, a similar temperature dependence of binding and a molecular weight of about 200,000 daltons. This site no longer possesses increased affinity in the presence of either GABA or anions indicating that those interactions depend on additional membrane-bound proteins or cellular reactions dependent on the binding of GABA. The cellular reactions enhanced by benzodiazepines are phospholipid methylation reaction which may play a role in altering membrane fluidity." @default.
