- W2088751410 abstract "1. The morphology of living amoebae during pinocytosis is described. These observations are correlated with observations on stained sections of amoebae fixed during pinocytosis. 2. Immediately before pinocytosis, wrinkling of the plasmalemma was observed. Wrinkling was followed by invagination of the plasmalemma which resulted in tunnels into the cytoplasm. These tunnels were then separated from the plasmalemma and were often broken up into droplets. 3. PAS and antibody staining procedures confirmed the observation that the pinocytosis vacuole membrane is derived from the plasmalemma. 4. All solutions of charged molecules which were tested, including many proteins and salts, were found to induce pinocytosis. Their activity was dependent to some extent on their concentration. 5. By fluorescence labeling techniques it was demonstrated that proteins are adsorbed to the plasmalemma. Salt solutions which caused pinocytosis were shown to cause a swelling of the outer coat of the plasmalemma. 6. A mechanism for pinocytosis is suggested which relates adsorption of charged solutes on the plasmalemma to a decrease in the tension and the structural rigidity of the membrane. The weakened membrane is drawn down into the cytoplasm by its “points of attachment” to the contractile plasmagel system. These invaginations become detached from the membrane and freed in the cytoplasm as pinocytosis droplets or vacuoles." @default.
