- W2088891434 abstract "われわれはこれまで,ラットに3%デキストラン硫酸ナトリウム(DSS)を自由飲水させることにより，体重の増加抑制，血便および貧血などの症状並びに大腸におけるびらんの形成，さらに小腸病変を欠くことなどの点でヒト潰瘍性大腸炎に類似した実験的潰瘍性大腸炎モデルが作製できることを確認した．また潰瘍性大腸炎発症動物のホルマリン固定大腸標本は，1%アルシアンブルーにより濃淡のある特徴的な青色に染色され，その濃青色領域は組織学的にびらんであることを明らかにした．今回，薬物の治療効果を評価する上で病態の程度が均等で，かつバラツキの小さい大腸炎モデルを作製するための実験条件について種々検討し，以下の結果を得た．1)ラットに3%DSSを連日自由飲水させ，飲水開始後に90%以上の個体に血便が観察された時点で選別した．2)選別基準は，(1)選別日を含めて少なくとも2日以上連続して血便が観察されること，(2)ヘモグロビン濃度が10g/d1以上であることおよび(3)選別当日の体重が，前日の体重に比べ20g以上減少していないこととした．3)選別基準を満足するラットを用い，選別日より1%DSSの自由飲水に切り替えることにより，血便の発現びらんの形成並びに赤血球数，ヘモグロビン濃度およびヘマトクリット値の低下，さらに生存率および体重などの面からみて，薬物の治療効果を判定できる適度な病態の維持が可能となった．4)選別日より1%DSSの自由飲水下でサラゾスルファピリジン(1日1回14日間経ロ投与)およびプレドニゾロン(1日1回7日間直腸内投与)を反復投与した場合，両薬物は潰瘍性大腸炎モデルに対して有意な治療効果を示した．以上の結果，本モデルを用いて1%DSSの自由飲水に切り替えた日より薬物投与を開始することにより，有用な潰瘍性大腸炎治療薬の評価が可能と考える．" @default.
