- W2089355841 abstract "Rationale: While the rate determining role of alveolar macrophages (AM) on phagocytosis and clearance of deposited micron-sized particles (MP) is well documented in the literature, there are clear indications that AM phagocytosis of deposited nanoparticles (NP) in the periphery is rather limited. Methods & Results: Based on our comprehensive and quantitative biokinetics studies up to six months after 1-h-short-term inhalation of 20nm iridium and titanium dioxide NP we confirmed that the phagocytised fraction of NP after 24 hours is about 25% of the peripherally deposited NP. The vast majority (˜75% of total lung burden) of NP is taken up in the epithelium and interstitial tissues of the lungs and is not available for broncho-alveolar lavage (BAL). However, very similar to the clearance kinetics of MP, NP are also cleared with a daily rate of about 0.02d-1 which is maintained by AM migration rate up the mucociliary escalator towards the larynx. In fact, at any time between three days and six months after inhalation all lavageable NP on the epithelial surface are phagocytised independent of the two NP materials used. As a consequence, NP had to re-appear gradually on the epithelial surface after their retention beyond the epithelium. It remains an open question when and where AM have phagocytised NP but it seems likely that this had already occurred at the interstitial retention sites since no free NP were observed in BAL fluids at any time during long-term retention. Conclusion: Hence, while the immediate defence capacity of AM is very limited directly after inhalation, the long-term clearance kinetics is not different to that of MP. Therefore, long-term clearance of NP in healthy adult rats is predominantly determined by the role of their AM." @default.
