- W2090165257 abstract "To the Editor: Similar to other analyses of articles published in several journals1Bardóczy Z. Györffy B. Kocsis I. Vásárhelyi B. Examination of Hardy-Weinberg equilibrium in papers of Kidney International: An underused tool.Kidney Int. 2004; 65: 1956-1958Abstract Full Text Full Text PDF PubMed Scopus (28) Google Scholar recently also reported an evaluation of manuscripts dealing with genetic polymorphisms and the underuse of calculations of Hardy-Weinberg equilibrium in Kidney International. Although this article has some merits, we wish to point out that the work of our group5Födinger M. Wölfl G. Fischer G. et al.Effect of MTHFR 677C>T on plasma total homocysteine levels in renal graft recipients.Kidney Int. 1999; 55: 1072-1080Abstract Full Text Full Text PDF PubMed Scopus (63) Google Scholar has been misreported and misinterpreted. In Table 1 of the article by Kocsis et al, our association study of a genetic polymorphism (MTHFR 677C>T) with total homocysteine (tHcy) plasma levels in kidney transplant patients5Födinger M. Wölfl G. Fischer G. et al.Effect of MTHFR 677C>T on plasma total homocysteine levels in renal graft recipients.Kidney Int. 1999; 55: 1072-1080Abstract Full Text Full Text PDF PubMed Scopus (63) Google Scholar has been referred to as having included 85 subjects showing a significant deviation of “MTFR-677” genotype distribution from the Hardy-Weinberg equilibrium (reference [19] of the article of Kocsis et al). Instead, we have applied the concept of Mendelian randomization in our study, and have included all 63 individuals that were homozygous for the MTHFR 677C>T from a total study population of 636 kidney transplant patients. These 63 homozygous subjects were matched with 63 heterozygous individuals (CT genotype), and with 63 patients homozygous for the wild-type alleles (CC genotype) for age, gender, estimated glomerular filtration rate, and body mass index, and were compared for biochemical markers of homocysteine metabolism. This study showed that MTHFR 677TT patients have significantly higher tHcy levels in contrast to patients with the CT or the CC genotype. Finally, multivariate analyses that controlled for important confounders confirmed this association. Thus, Kocsis et al should more carefully read, analyze, and interpret the data reported in the literature before concluding that deviations from Hardy-Weinberg equilibrium need more consideration in gene-association studies." @default.
