- W2090985893 abstract "Gram-negative bacteria are responsible for a number of infections and accounts for a high morbidity and mortality, influencing the outcome of surgical procedures. Endotoxin is known to play an important role in sepsis and related syndromes. Endotoxemia causes release of eicosanoids and production of free radicals, responsible for the development of endotoxemic shock. The pathophysiologic characteristics of this severe situation are capillary leakage followed by hemodynamic instability and disturbed microcirculation causing multiorgan failure and disseminated intravascular coagulation. However, data on the development of hemostatic disorders in sepsis are conflicting, and there is little information about the influence of endotoxin on primary hemostasis. Since thrombocytosis and thrombocytopenia are common signs of septic situations, the involvement of platelets seems evident. Prior investigations on this topic described direct endotoxic lipid A stimulation of human platelets by activation of protein kinase C [ 1 Grabarek J. Timmons S. Hawiger J. Modulation of human platelet protein kinase C by endotoxic lipid A. J Clin Invest. 1988; 82: 964-971 Crossref PubMed Scopus (25) Google Scholar , 2 Romano M. Hawiger J. Interaction of endotoxic lipid A and lipid X with purified human platelet protein kinase C. Lipopolysaccharide induces exposure of fibrinogen receptors on human platelets. Chin Med Sci J. 1995; 10: 73-77 PubMed Google Scholar ]. Other authors observed this phenomenon only in whole blood [ 4 Nystrom M.L. Barradas M.A. Jeremy J.Y. Mikhailidis D.P. Platelet shape change in whole blood Differential effects of endotoxin. Thromb Haemost. 1994; 71: 646-650 PubMed Google Scholar ] and proposed it to be mediated by activated monocytes and/or neutrophils [ 5 Csako G. Suba E.A. Elin R.J. Endotoxin-induced platelet activation in human whole blood in vitro. Thromb Haemost. 1988; 59: 378-382 PubMed Google Scholar ]." @default.
