- W2091571738 abstract "OBJECTIVE: To compare laboratory and clinical outcomes of SIN vs SEQ using sibling oocytes/embryos from day of retrieval to day 6. Also, to better determine the impact of these two types of culture media systems on pregnancy rates (PR), comparisons were made between groups of patients where all oocytes/embryos were placed in either SIN or SEQ. DESIGN: Retrospective study. MATERIALS AND METHODS: All IVF/ICSI patients from June 2007 to January 2008 were analyzed in this study. Patients were separated into one of three groups: a) sibling oocytes split between SIN or SEQ (N=27), b) all oocytes in SEQ (N=43), c) all oocytes in SIN (N=43). SEQ used was from SAGE and it consisted of Fertilization media (day 0-1), Cleavage media (day 1- 3) and Blastocyst media (day 3-6). SIN was Global and eggs/embryos were transferred to fresh drops of media on days 0, 1, 3 and 5. All media was supplemented with 10% Synthetic Protein Substitute and was overlaid with oil. Incubators had an atmosphere of 6.5% CO2 in room air. RESULTS: There were no significant differences in stimulation protocol, age, baseline serum FSH levels or number of embryos transferred between groups. Comparison of sibling embryo delopment demonstrated a significant increase in the number of embryos with > 6 cells on day 3, total blastocyst formation and blastocysts cryopreserved by day 6 in SIN over SEQ. This improved embryo development was also demonstrated when the sibling embryo data was combined with the embryo data from the cases where all the eggs were placed in SEQ or SIN. This also allowed for a comparison of PR. For this comparison, split cases where all the embryos transferred were from only SEQ or SIN were included.
