- W2093204530 abstract "Chromosome aberration and sister chromatid exchange (SCE) frequencies were determined in both lymphocytes and Ph1-positive cells from chronic myeloid leukaemia (CML) patients before and after treatment with busulphan (BUS). For blood with a high leucocyte count, lymphocyte separation techniques were used to increase the percentage of T lymphocytes in cultures, thereby improving the mitotic yield. Increases in the incidence of chromosomes aberrations in lymphocytes from treated patients could persist for more than 6 months after therapy. However, single high doses of BUS induced a transient increase in SCE levels in patients' lymphocytes, which returned to control levels within a few weeks. Only when samples were taken from patients within 2 days after receiving a large dose of BUS were increases in chromosomes aberration and SCE frequencies detected in Ph1-positve cells. The distribution of aberrations in G-banded metaphases of lymphocytes from patients treated with BUS were non-random and a number of chromosome bands which were frequent sites for aberrations for in vitro BUS exposure were also found to be particularly susceptible following in vivo BUS exposure." @default.
