- W2093385483 abstract "This paper reports the conversion of 14C-labeled geranylgeranyl pyrophosphate to phytoene by a soluble enzyme system obtained from tomato plastids. Proof of the identity and purity of the 14C-geranylgeranyl pyrophosphate is presented in the preceding paper and some additional information is presented in this paper. Proof of the incorporation of radioactivity into phytoene was obtained through a demonstration of (1) the coincidence of radioactivity with phytoene on chromatography on alumina, and (2) the coincidence of radioactivity and mass of hydrogenated phytoene (lycopersane) on gasliquid chromatography. The incorporation of radioactive substrate into phytoene was maximal at pH 7.6 to 7.8 and at 25 ° C. The synthesis of phytoene from geranylgeranyl pyrophosphate was also dependent upon the presence of Mg++ and a sulfhydryl reagent (dithiothreitol). Presumably the latter compound functions by maintaining sulfhydryl groups of the enzyme in the reduced state." @default.
