- W2094621088 abstract "Abstract Determination of the absorbed dose is essential to study the biological effects of ionizing radiation. Dosimetry requires the knowledge of cell thickness in the same conditions as used in the designed experiments. This can be obtained by measuring charged beam energy loss in the sample. We used an accelerated α-particle beam and CR-39 track-etch detectors or silicon surface barrier detectors to measure the energy. In the first case, cells were grown on CR-39 plastics, in the latter cells were grown on a thin mylar foil placed in front of the silicon detector. Cell thickness was obtained as the difference of ranges R (E)− R ( E ′), where E ′ and E are the measured energies with and without the cell monolayer in front of the detector, respectively. The measured thickness of C3H 10T1/2 murine fibroblasts growing as a monolayer on CR-39 plastics was 3.3 ± 2.0 μm, and 3.6 ± 1.9 μm when grown on mylar films. These results are compared with those obtained in the same cell line by microscopy in other laboratories. Nuclear methods provide reasonable estimates of the cell thickness on living cells, in the same conditions adopted for radiobiological experiments, and provide information on the fluctuations in the monolayer thickness.
