- W2095160745 abstract "Thrombosis is one of the major causes of human death worldwide. Identification of the cellular and molecular mechanisms leading to thrombus formation is thus crucial for the understanding of the thrombotic process. To examine thrombus formation in a living mouse, new technologies have been developed. Digital intravital microscopy allows to visualize the development of thrombosis and generation of fibrin in real-time within living animal in a physiological context. This specific system allowed the identification of new cellular partners involved in platelet adhesion and activation. Furthermore, it improved, especially, the knowledge of the early phase of thrombus formation and fibrin generation in vivo. Until now, platelets used to be considered the sole central player in thrombus generation. However, recently, it has been demonstrated that leukocytes, particularly neutrophils, play a crucial role in the activation of the blood coagulation cascade leading to thrombosis. In this review, we summarized the mechanisms leading to thrombus formation in the microcirculation according to the method of injury in mice with a special focus on the new identified roles of neutrophils in this process. La thrombose est l’une des causes majeures de mortalité dans le monde chez l’Homme. L’identification des mécanismes cellulaires et moléculaires impliqués dans la formation d’un thrombus est donc cruciale pour la compréhension du processus thrombotique. De nouvelles technologies ont été développées afin de modéliser le processus de thrombose in vivo. La microscopie digitale intravitale permet de visualiser en temps réel le développement d’un thrombus et la génération de fibrine dans l’animal vivant dans un contexte physiologique. Ce système spécifique permet l’identification de nouveaux partenaires impliqués dans l’adhésion et l’activation plaquettaire. De plus, la microscopie digitale intravitale en temps réel permet l’identification des différents partenaires impliqués dans les phases précoces de la formation d’un thrombus in vivo. Jusqu’à maintenant, les plaquettes étaient considérées comme les seuls partenaires majeurs jouant un rôle dans la formation d’un thrombus. Récemment, il a été démontré que les leucocytes, en particulier les neutrophiles, jouaient un rôle important dans l’activation de la cascade de coagulation menant à la formation d’un thrombus plaquettaire. Cette revue présente les mécanismes mis en jeu dans la formation d’un thrombus en fonction des différents modèles de blessure dans la microcirculation de la souris et décrit spécifiquement le rôle des neutrophiles dans la phase précoce de développement du thrombus." @default.
