- W209720870 abstract "To detect the appropriate concentration of drug used in the drug-susceptibility test(DST) for Ethambutol-resistant isolates of Mycobacterium tuberculosis,5 concentration gradients(2.4 μg/L.2.0 μg/L,1.6 μg/L,1.2 μg/L,0.8 μg/L)of drug were used to detect the Ethambutol susceptibility and the rates of drug resistance were compared.The statistical methods used in the present study included chi-square test,logarithmic fit curve,established equations and estimated parameters,and the P value standard adopted was 0.05.It was demonstrated that the detection rate of Ethambutol-resistance of 26 sensitive strains under 5 concentration gradients of drug was 0%,while those of 28 mutant strains under these concentration gradients of drug and bacterial concentrations of 10-3 mμg/L and 10-5 mμg/L were 0%,0%,82.1%,89.3%,96.4% as well as 0%,0%,25%,42.9%,53.6% respectively.There was significant difference in the detection rate of the resistant strains between drug concentrations of 2.0 μμg/L and 1.6 μμg/L rather than those of 1.6 μg/L and 1.2 μg/L with definitely statistical significance(P0.05).As a whole,the detection rate of resistant strains in the Ethambutol concentration of 1.6 μg/L appeared to be higher.Meanwhile,in the logarithmic fit curve,the parameter containing the R square was0.774 and the fitted equation was y=0.923+(-0.974) Inx,indicating that the selected concentrations were accordant with the practical situation.It is concluded that the traditional concentration of drug used in the susceptibility test for Ethambutol appears to be rather higher and the concentration of 1.6 μg/L actually can increase the detection rate of resistant strains of M.tuberculosis in DST.An accurate concentration of Ethambutol between 1.6 μg/L and 2.0 μg/L can accurately reflect the true situation of drug-resistance in combination with the laboratory and clinical investigation." @default.
