The main problem is the relatively weak forces involved in the native conformation of these proteins and so making them quite labile. Furthermore, their susceptibility to proteolytic enzymes in the gut makes oral administration quite challenging. While various routes like, ocular, transdermal, nasal and buccal have been tried, none of these routes has proved to be a potential alternative to the invasive injection. However, various studies have been performed on the formulation of these proteins as aerosols for pulmonary delivery and promising results have been obtained.
