- W2100367065 abstract "Deposition of amyloid b peptide (Aβ) into the brain is one of molecular events in the brain of Alzheimer's disease (AD) associated with cognitive impairment. Epidemiological studies suggest that dietary intake of fishes enriched omega-3 fatty acids such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) reduces the risk of AD. We investigated the effects of prescription pre-administration of omega-3 fatty acids on the impairment of learning ability in AD model rats infused with Aβ1-40 peptide into the cerebral ventricle and the Aβ1-40 levels in the nonionic detergent Triton 100X-insoluble membrane fractions (DIFs) of the cerebral cortex. Learning-related behavior was assessed using an 8-arm radial maze. The levels of Aβ1-40 were analyzed with an ELISA kit (Wako Pure Chemical Industries, Ltd., Osaka, Japan). Pre-administration of TAK-085 (a highly purified and concentrated omega-3 fatty acids containing EPA ethyl ester, and DHA ethyl ester) at 300 mg/kg/day for 12 wk to the Aβ1-40-infused rats significantly reduced the increase in the number of reference and working memory errors in the maze task in the Aβ-infused rats with a concomitant increase in the cortico-hippocampal EPA and DHA, suggesting the preventive effect of TAK-085 from the impairment of learning ability in AD model rats. The infusion with Aβ1-40 peptide increased the levels of Aβ1-40 peptide in the DIFs and also the levels of lipid peroxide (LPO) and reactive oxygen species (ROS) of the cerebral cortex. The pre-administration of TAK-085 suppressed the increases in the levels of Ab1-40 peptide in the DIFs, and the LPO and ROS levels in the cerebral cortex of Ab-infused rats. The present studies suggest that long-term administration of TAK-085 could be used as a possible therapeutic agent for protecting learning deficiencies and deposition of Aβ due to Alzheimer's disease." @default.
