- W2103165050 abstract "Twoa-tubulin genesfromthebudding yeastSaccharomyces cerevisiae were identified andcloned by cross-species DNA homology. Nucleotide sequencing studies revealed thatthetwogenes, namedTUB)and TUB3,encoded gene products of447and445aminoacids, respectively, thatare highly homologous to oe-tubulins fromother species. Comparison ofthesequencesofthetwogenesrevealed a 19%divergence between thenucleotide sequencesanda 10%divergence between theaminoacid sequences.Eachgenehada single intervening sequence, located atan identical position incodon9.Cell fractionation studies showedthat both geneproducts werepresent inyeast microtubules. These twogenes, along withtheTUB21-tubulin gene, probably encode theentire complement oftubulin inbudding yeastcells. Theprocessofcell division hasbeenstudied extensively inthebudding yeastSaccharomyces cerevisiae andthe fission yeastSchizosaccharomyces pombe.A particularly useful technique hasbeentheisolation ofconditional lethal mutantsthatarrest atspecific morphological stages ofthe cell cycle (23, 35,38,39,47,56,70). Thenormal function of themutantgene product insuchcellcycle mutantshas, unfortunately, beendiscovered inrelatively fewinstances. Todefine therole ofthese geneproducts inthemechanism ofcell division, itwill beessential tocombine thegenetic approach withbiochemical andmorphological analysis. Yeastshavebecomefavorite organisms forsuchstudies because ofeaseofgrowth andmanipulation, because ofthe sophisticated recombinant DNA andclassic genetic techniques that havebeendeveloped (11, 61), andbecause ofthe successful application ofsuchtechniques aselectron microscopy andimmunofluorescence" @default.
