- W2103267052 abstract "incell cultures derived fromthesame specimens andprotein assayson theculture supernatants. Placental protein 14(PP14) was produced bynormalpremenopausal epithelium butnotbythe post-menopausal ormalignant endometrial epithelium. Incontrast, placental alkaline phosphatase (PLAP) was produced byendometrial cancersandtheendometrial adenocarcinoma-derived cell line Ishikawa, butnotby thenormal endometrial epithelium. Othermarkers suchasCA-125, which was produced bybothnormaland malignant endometrium butnotbythecell line, andhumanchorionic gonadotrophin (P-hCG), whichwas produced byIshikawa cells butnotbyany ofthefresh tissues, were less cancerspecific. Placental alkaline phosphatase isadirect product ofendometrial cancersthat canbereadily assayed inserumusing this two-site assaytotestitsclinical usefulness inmonitoring patients atriskforendometrial cancer. Tumourmarkersforendometrial cancerhavereceived relatively little attention because ofthehighrateofearly detection andcureinmostwomenwhopresent withpostmenopausal bleeding. However, interest hasbeenaroused by theincreasing needtomonitor womenwhoarereceiving long-term tamoxifen (Uzeily etal., 1993) andthose apparently curedoftheir endometrial cancer, whoyetwishto receive thebenefits ofhormone replacement therapy (Creasmanetal., 1991). Placental protein 14(PP14) (Bolton etal., 1983) isoneof themajor proteins secreted bythenormal endometrium and thedecidua inearly pregnancy (Bell etal., 1985; Julkunen et al., 1985). Placental alkaline phosphatase (PLAP), anisoenzymeofthealkaline phosphatase group(Fishman etal., 1968), originally isolated fromhumantrophoblast, isalso expressed inother non-malignant humantissues (Goldstein et al., 1982; McLaughlin etal., 1984). PLAPisfoundtobe elevated intheseraofsomepatients withcancers, particularly those ofthereproductive tract (Nathanson & Fishman1971; Wahrenetal., 1979). Thebetasubunit ofhuman chorionic gonadotrophin (P-hCG), anormal product ofthe trophoblast, andCA-125(Bast etal., 1981) arebothwellcharacterised oncofetal antigens (Braunstein, 1983) found to besynthesised bymanynormal tissues andepithelial carcinomas ofthereproductive tract. We haveestablished aninvitro system fortheculture of normal andmalignant endometrial epithelium. Thedifferential expression ofmarkers hasbeencharacterised infrozen tissue sections, primary cell cultures andinanestablished endometrial cell line, inordertodemonstrate their specific production bythecancers asa preliminary toevaluating their useasclinical markers." @default.
