- W2105100109 abstract "It is known that human blastocysts are able to secrete in vitro a wide repertoire of growth factors and cytokines that can be involved in the mechanisms that allow the embryo to create their own inmmunoprovilege site and therefore to avoid early rejection during implantation. The aim of this study was to investigate whether human blastocysts are able to regulate the in vitro the expression of two early activation markers (CD25 and CD69) on peripheral blood mononuclear cells (PBMC). Prospective study. 14 patients participating on our ovum donation programme and 33 human blastocysts were included in this study. We divided the human embryos into Group A comprised by 10 blastocysts able to implant and group B comprised by 23 blastocysts that did not implant. Human embryos were grown in 50 ml drop of in house HTF till day 3 and from day 6 onwards in CCM (Vitrolife). After embryo transfers, blastocyst conditioned media (BCM) was stored at −20°C. PBMC from fertile women were isolated on day LH + 7.2 × 106/ml phytohemagglutinin-activated PBMC were cultured for 72 hours in presence of 20% BCM or 20% of CCM for the negative control. All experiments were done in duplicates. Using the labeled strept-avidin-biotin immunoperoxidase technique, we examined CD25 and CD69 antigen expression. 200 cells were counted at 40x in each experiment. Both antigen expression were normalized by the results generated by the negative control. Values greater than 1 showed a positive relative increase while values lower than 1 expressed a negative regulation. Statistical analysis: T-Student test. P values < 0.05 were considered statistically significant. Significant decrease in both CD25 and CD69 relative expression was induced by BCM generated from blastocysts that successfully implanted in comparison of blastocysts that failed to implant (0.85 vs. 1.15 and 0.93 vs. 1.05 respectively). Human blastocysts with successful implantation potential were able to down regulate the immune responses from activated PBMC, it may show that the embryo need to have an active role in order to guarantee a proper immune environment to allow implantation." @default.
