- W2106021714 abstract "An in vivo spectrophotometric technique for estimating photosynthetic pigments of intact phytoplankton was examined by combining phytoplankton cultures, chemical analyses of pigments and computerized optical measurements. Chlorophyll a, b and c concentrations of Anabaena flos-aquae, Chlorella sp. No. 227, Cryptomonas ovata var. palustris and Prorocentrum mariae-lebouriae were estimated. The absorbance spectra log (1/T) from 300 to 800 nm of mixed phytoplankton populations collected on filters were recorded with a computerized spectrophotometer. The regression used a single-term second derivative of the log (1/T) data to predict a given concentration from absorbance and chemical measurements with high accuracy. The computer test showed 553·5 nm to be the best wavelength for estimating chlorophyll a, 642 nm for chlorophyll b and 606 nm for chlorophyll c concentrations. The correlations between chemical analyses and absorbance values were: r=0·996 for chlorophyll a varying from 25 to 40 µg per filter, r=0·99..." @default.
