- W2108850579 abstract "The endometrium of women with polycystic ovary syndrome (PCOS) is associated with progesterone (P4) resistance and abnormal steroid receptor/co-activator profile which likely contribute to increased miscarriage rate and risk of endometrial cancer in these women. Whether there is an inflammatory component in PCOS endometrium that relates to endometrial dysfunction is not known. We hypothesized that endometrial stromal fibroblasts (eSF) show aberrant steroid hormone responsiveness and production of pro-inflammatory cytokines in PCOS. In vitro study. Cultures of eSFPCOS (n=8) and eSFcontrol (n=5) were treated with vehicle, estradiol (E2, 10nM) or E2P4 (10nM/1uM) for 14 days. Decidualization of eSF was confirmed by insulin-like growth factor-binding protein 1 (IGFBP1) ELISA. Interleukin (IL) 4/6/8, granulocyte macrophage colony stimulating factor (GM-CSF), macrophage chemoattractant protein (MCP) 1/3, CX3CL1 and CCL5 were measured in the conditioned media by Luminex. ANOVA with pre-planned contrasts was used for statistical analysis. Hormonal challenge with E2P4 to induce decidualization revealed two distinct subsets of eSFPCOS. Six eSFPCOS and all eSFcontrol cultures showed normal decidualization response to E2P4, whereas 2 eSFPCOS cultures failed to decidualize. These subsets of eSFPCOS cultures also behaved differently with respect to cytokine secretion both in the presence and absence of E2 and E2P4. The cytokine response patterns to E2 or E2P4 in decidualizing eSFPCOS were similar to controls, whereas non-decidualizing eSFPCOS cultures showed increased (P<0.05) IL4/6 and MCP1 levels in all treatment groups compared to eSFcontrol or decidualizing eSFPCOS. In some women with PCOS, eSFs show aberrant response to E2P4 and elevated cytokine secretion which may contribute endometrial dysfunction in these women." @default.
