It begins by describing in schematic terms the current paradigm in drug research and development, and goes on to explain why and how ADME-Tox has gained an ever increasing significance in the screening and assessment of lead candidates, leads, and clinical candidates. The chapter then draws a parallel between the modern ADMET paradigm and the rationale underlying the organization of the volume. Because the ultimate ‘target’ of drug R&D is the human patient, the volume begins with the clinical pharmacokinetic criteria good candidates are expected to fulfil, and with the biochemical bases of drug metabolism and toxification/detoxification. The other parts of the volume deal successively with screening and evaluation tools ranging from in vitro ADMET bioassays to ADME-relevant physicochemical techniques to virtual ADMET experimentation using in silico algorithms." @default.
