Semen cryopreservation procedures have been evolving over the past 50 years. However, despite continued advances in the development of newer extenders and cryoprotectants, the post-thaw motility of bovine sperm remains at around 50%. This is because semen cryopreservation involves several general steps, of which dilution, cryoprotection, cooling and freezing, storage, and thawing can affect sperm structure and function. There are currently two standard protocols approved for freezing bovine semen commercially by the Certified Semen Services Inc. (CSS). The standard CSS protocol or the two-step method is the most common method utilized in the United States, whereas the one-step method (also known as the alternative CSS protocol) is more popular in Europe.
