- W212305460 abstract "Multiple genetic alterations have been implicated in the development of colorectal carcinoma including involvement of tumor suppressor genes, oncogenes and mutator genes, which are characterized by the presence of microsatellite instability (MIN). Conventional cytogenetic studies have demonstrated both frequent clonal losses (chromosomes 4, 8, 10, 14, 17, 18, X and Y) and clonal gains (chromosomes 7 and 13). In this study, fluorescence in situ hybridization (FISH) using chromosome specific directly-labelled {alpha} satellite probes was used to evaluate aneuploidy (gain or loss) of chromosomes 4, 7, 8, 10, 12, 17, 18 and X in isolated nuclei from paraffin-embedded tissue from 25 proximal and 25 distal colorectal tumors. Numeric abnormalities were present in 46/50 tumors. Chromosome 7 was most commonly gained (31/50 cases) and chromosome 18 was most commonly lost (20/50 cases). The average number of abnormal chromosomes per case was similar in the proximal (4.8/8 chromosomes) and distal (5.5/8 chromosomes) tumors. However, a significantly higher average chromosomal gain was observed in the distal tumors (3.8/8) compared to proximal tumors (2.0/8, p=0.01). In contrast, the proximal tumors had a higher, but not statistically significant, average loss of chromosomes (2.8/8) compared to the distal (1.7/8) tumors. Different patterns of chromosomal gains and lossesmore » were also observed between MIN+ (10) and MIN- (15) proximal tumors. Loss of chromosomes was significantly higher in the MIN+ (average 4.3/8) than in the MIN- (average 1.9/8) group of tumors (p=0.006). In conclusion, aneusomy involving multiple chromosomes is a common event in colorectal tumors. Different patterns of aneusomy are present in proximal and distal tumors and in tumors with MIN. As assessed by FISH analyses, distal tumors have significant gain of chromosomes compared to proximal tumors.
