The magnitude of class II-restricted T-cell responses is measured directly in vitro (T-cell proliferation), indirectly in vitro (lymphokine production, help to antibody production by B cells), or indirectly in vivo (delayed hypersensitivity, serum antibody titers). After the elaborate description of these Ir gene phenomena, it has since become overwhelmingly clear that cytotoxic T-cell (CTL) responses to various antigens are also MHC Ir gene controlled. In this instance the responsible Ir gene productions are both the class I and class II MHC molecules1–5. This creates additional complexity of MHC regulation, because now the response is regulated directly by class I MHC molecules at the level of class I-restricted CTL precursor (CTLp) activation as well as at the level of class II-restricted TH cells promoting optimal differentiation and expansion of CTL. Indeed, although helper-independent CTL have been described6, most CTL responses are clearly augmented by TH cells. In many instances the CTL response is even completely dependent on proper class II-specific (or -restricted) TH cell activation.
