Gas liquid chromatography (GLC) was preferred for drug monitoring studies, while atomic absorption spectroscopy (AAS) was the method of choice in detecting disulfiram overdosage by urinalysis. GLC was proved to be the most sensitive and reliable technique for quantitation of free NaDEDC in serum. This compound is readily derivatized into its dichlorobenzylester and separated by GLC on a DC-200 stationery phase after extraction with n-hexane. Soidum dimethyldithiocarbamate (NaDMDC) is added as an internal standard before the extraction procedure. Linearity started from a 50 µg/L concentration with ECD and from 500 µg/L with NFID in serum. For rapid analysis of 2-ml urine samples after acute intoxication or overdosage, NaDEDC was determined by treating the urine with a buffer at pH 4 and an excess of Cu2+. The Cu chelate produced in this way is extracted with CCl4 and subjected to Flame Atomic Absorption Spectroscopy (F.A.A.S.), which is recommended for urinary NaDEDC levels lower than 5 ng/L. Quantitation was performed by an external standard procedure with a calibration curve set up by an inorganic copper standard series and by internal standard addition." @default.
