- W2130708045 abstract "RATIONALE: Nitrite and nitrate are nitrogen oxides derived from several sources in the lung and digestive tract. Increased concentrations of these metabolites have been reported in exhaled breath condensates (EBC) from patients with pulmonary disorders and in some cases correlated with changes in exhaled nitric oxide (eNO). The aim of the present study was to determine whether nitrite and nitrate could be detected in EBC from patients with mild persistent asthma and investigate the effect of treatment with a combination of an inhaled corticosteroid and long acting beta-2 agonist on these metabolites. METHODS: Ten healthy volunteers and 18 mild persistent, steroid-naïve asthmatics (age 22-62 years, FEV-1 > 70%) were enrolled. In this open label study, the asthmatic group was treated with FSC 100/50, 1 inhalation BID, for 4 weeks. eNO and EBC nitrite/nitrate concentrations were measured before and after 4-week of therapy. RESULTS: eNO was increased in the asthmatic group (74 (SD=37) ppb; p<0.001) compared to controls (24 (SD=7) ppb). Nitrite concentrations from asthmatics were significantly increased (2.3 (SD=2.5) μM; p<0.05) compared to controls (1.4 (SD=0.5) μM). This could not be accounted for by a change in EBC pH. After therapy, eNO fell from 74 (SD=37) ppb to 34 (SD=15) ppb (p<0.001) but there was no statistically significant changes in either nitrite or nitrate. CONCLUSION: EBC nitrite is elevated in asthmatic patients. The lack of correlation between eNO and nitrite and absence of significant changes in nitrite and nitrate concentrations upon treatment would presumably reflect different metabolic origins for nitrite, nitrate, and eNO." @default.
