- W2133049038 abstract "In order to further evaluate the immunosuppressive effects of cyclophosphamide, the present study examined the activities of drug therapy on the immune response to influenza A vaccine. Twelve rheumatoid patients receiving cyclophosphamide (0.5 to 1.5 mg. per kilogram per day) were studied prior to and 3 weeks after immunization with bivalent influenza vaccine; controls consisted of a group of 13 rheumatoid arthritis patients not receiving cytotoxic agents. Both groups showed comparable increases in HAI antibody titers. The increase in the control group was 5.0 + 1 .l (l/log, titer) while the average increment in the drug-treated patients was 3.9 * 1 .l. Further, there was no qualitative difference in the type of antibodies present at the time of testing; in neither group was the titer altered by reduction with P-mercaptoethanol. In contrast, in vitro lymphoproliferative responses to influenza antigen were markedly restricted in the patients treated with cyclophosphamide. In these patients, the average H3Tdr incorporation was reduced by 85 per cent in the preimmunization sample and 79 per cent in those samples obtained 3 weeks after immunization. A similar reduction in isotope uptake was noted in cultures stimulated with two nonspecific mitogens, PHA (53 per cent reduction) and pokeweed (61 per cent reduction) and in the activity in unstimulated cultures. These results indicate that prolonged therapy with cyclophosphamide does not acutely impair the elaboration of 7s antibodies in an anamnestic type response but markedly limits the ability of immunologically responsive lymphocytes to undergo in vitro proliferative expansion." @default.
