- W2134691036 abstract "The advancing edge of Walker 256 carcinoma was examined by light and electron microscopy in tumors growing in rat thigh muscle and on the cheek pouch of the golden hamster. In vivo observations were made on the microcirculation of Walker 256 carcinoma implanted on the hamster cheek using Sanders-Shubik chambers. Capillary fragility was pronounced at the leading edge of the tumor and for some little distance ahead of it. Tumor growth within optical chambers was sufficient to cause an increase of tissue tension resulting in occlusion of the microcirculation of the central area of the tumor. This became apparent when the pressure was released by removal of the base plate in a controlled fashion. At the advancing edge there was the ingrowth of capillary sprouts from the host's pre-existing capillary network and these were associated with mitotic figures in endothelial cells both in small venules and capillaries. Cross connections between the capillary sprouts developed and a fine microcirculation was formed. The early microvasculature of this tumor showed intracellular endothelial fenestrations and saccular endothelial processes. The cytoplasm of the newly formed capillaries was occasionally humped into high ridges. The fenestrations which separated these cytoplasmic ridges were not evenly distributed and were closed by a diaphragm which had a thickness equal to that of the plasma membrane of the cell.
