Primarily, these oral agents have been used as premedicants to provide sedation and/or analgesia to complement the induction of anesthesia via intravenous or inhalation routes. However, seldom has the oral route been used as the sole means for rendering a patient unresponsive to surgical stimulus. Much of this is related to the fact that when compared with parenteral drug administration, the oral route does not offer the same efficacy and safety due to multiple issues relating to drug uptake, duration, and the inability to titrate to effect. Drugs administered per oral (PO) are subject to hepatic metabolism before entering the systemic circulation. A drug’s first-pass metabolism combined with gastric degradation and varied rate of absorption often makes for unpredictable drug bioavailability." @default.
