Titration of a further 7–9 SH groups by either reagent is less rapid and is accompanied by inactivation. A further 45–50 SH groups were titrated by both reagents after exposure to 6 m or 5.2 m GCl. Behavior of NEM-inactivated urease differed from that of DTNB-inactivated urease during isoelectric focusing. DTNB titration in 8 m urea solution did not indicate that significantly more SH groups were exposed under these conditions, but NEM reacted with about 50 SH groups. No evidence was found for the presence of disulfide bonds in the molecule. The number of SH groups titratable in GCl solution or in peptic digests was equal to the number of 12-cystine residues from previously published amino acid analysis." @default.
