- W2136276506 abstract "Phytohemagglutinin (PHA) has been shown to stimulate both mitoses and the formation of blast-like cells in cultures of normal human leukocytes (1). The blast-like cells have been studied in the living state in this laboratory (2). In the absence of knowledge of their character and their potentialities, these cells were called PHA-cells. The previous studies showed that PHA produced PHA-cells in purified blood cell suspensions consisting almost entirely of lymphocytes, inhibited the transformation of monocytes to macrophages, caused the clumping of leukocytes, and failed to stimulate the formation of PHA-cells in cultures of blood cells from the rat and from most patients with chronic lymphocytic leukemia. It seemed difficult to conceive that phytohemagglutinin is a unique reagent and that the PHA-cell can be produced only by this reagent. Furthermore, even in the absence of PHA, an occasional suspension, derived from normal blood, was seen in this laboratory to develop a few cells morphologically similar to PHA-cells (see Figure 6, in reference 3). Consideration was, therefore, given to other agents that might produce PHA-like reactions on blood cells. Tuberculin purified protein derivative (PPD) was accordingly tested and parallel experiments were made with PHA. The present paper describes the similarities and differences which were observed in the biologic activity of PPD and PHA on blood cells in culture." @default.
