- W2137934071 abstract "Following the introduction of the cluster of differentiation (CD) nomenclature in 1980, flow cytometry became the method of choice for lymphocyte subset enumeration as well as immunophenotyping leukemias and lymphomas. After the description and characterization of the CD34 Ag, various methods for enumeration of hematopoietic stem and progenitor cells (HSC) by flow cytometry were developed. The introduction of flow cytometric HSC enumeration was clinically highly useful for planning apheresis sessions to collect peripheral blood HSC and assessing the engraftment potential of HSC grafts. With the increasing use of flow cytometry in clinical programs, it soon became evident that there was a need to harmonize the methods used for cell analysis among laboratories. Therefore, various guidelines for assays, such as CD4+ T-cell enumeration in HIV-infected patients, leukemia/lymphoma immunophenotyping, CD34+ cell enumeration and screening for the HLA-B27, were developed. Along with the development of these guidelines, programs for external quality assurance were developed, aimed at reducing the variability in assay outcomes between laboratories and improving the quality of assay performance at individual laboratories [ 1.
