- W2143587507 abstract "Alzheimer's Disease (AD) is the most common cause of dementia in the Western world. Cholinesterase inhibitors (ChEIs) have been shown to enhance cognitive functioning in patients with AD. The use of ChEIs and their maximum dose is limited by side effects, such as nausea, vomiting and diarrhoea. These side effects are caused by stimulation of peripheral acetylcholine receptors. Memogain is a prodrug of galantamine that preferentially enters the CNS where it is cleaved to active galantamine. This mechanism is supposed to lead to lower levels of galantamine in the peripheral circulation and higher levels in the brain, with the medical benefits expected to be fewer side effects and larger cognitive enhancement. At present we are addressing these effects in phase-I clinical settings. This is a double-blind, double-dummy, randomized, placebo- and reference- controlled, dose-escalation study. ln the first two cohorts, healthy young men received Memogain 5,5mg, 11mg or placebo. Several computerized tests for memory, attention, Stroop test, visual analogy scales (VAS), eye movements and pharmaco-EEG were performed repeatedly after drug administration. These results are presented in this abstract. In cohort 3, 4 and 5 healthy elderly subjects will receive Memogain 22mg, 33mg or 44mg or galantamine 16mg or 24mg or donepezil 10mg. These results will be available in April 2014. Memogain nasal spray 5.5mg and 11mg was well tolerated. A difference between Memogain 5.5mg and Memogain 11mg versus placebo was found for adaptive tracking, a very sensitive measure for attention (respectively 1.96 (CI -0.86 - 4.80) and 3.48 (CI 0.54 - 6.42); figure 1), N-back, a test for working memory (respecitvely 0.09 (CI -0.06 - 0.24) and 0.12 (CI -0.02 - 0.27); figure 2) and on the visual verbal learning test, a test for learning and retrieval of information (figure 3 and 4). No differences were found on Stroop test, eye movements, VAS and pharmaco-EEG. The administered doses of Memogain were well tolerated. A dose dependent positive effect on attention and several aspects of memory was observed. Memogain may be a promising treatment for symptoms of Alzheimer's Disease. Results of the higher doses and comparison with galantamine and donepezil may confirm these results and will be available in April 2014." @default.
