- W2153192005 abstract "The presence of purified endo-α-1,3-glucanase reduced water-insoluble glucan (ISG) production and increased soluble glucan (SG) production from sucrose by the cell-free glucosyl-transferases of Strep. mutans AHT. Native ISG comprised dextranase-susceptible glucan as single-stranded fibrils and electron-dense globular masses, as well as dextranase-resistant glucans as double-stranded fibrils. The fibrillar components formed a meshwork in which the globular components were surrounded by layers of the double-stranded fibrils. The globular masses appeared to be compact arrangements of the single-stranded fibrils. Compared to native ISG, ISG synthesized in the presence of α-1,3-glucanase lost the bulk of the dextranase-susceptible, single-stranded, fibrillar component and consisted mainly of parallel aggregates of a shorter form of the dextranase-resistant, double-stranded fibrillar component. The SG produced in the presence of α-1,3-glucanase contained single-stranded fibrils and electron-dense globular components and showed little morphological alteration compared to native SG. These observations suggest that the single-stranded fibrillar component of ISG is an alternative state of the single-stranded fibril constituting SG. Both the ISG and the SG produced in the presence of α-1,3-glucanase formed smaller aggregates compared to their respective native control glucans. The presence of these small glucan aggregates may explain the enhanced agglutination of Strep, mutans cells by these glucans. These observations support the mechanisms of inhibition of ISG synthesis by α-1,3-glucanase proposed in chemical studies." @default.
