Birinci grubu fizyolojik tuzlu su uygulanan fareler (KFTS grubu, kontrol), ikinci, ucuncu, dorduncu ve besinci grubu sirasiyla 100 (L100), 250 (L250), 500 (L500) ve 1000 (L1000) µg/kg/gun dozlarinda rekombinant fare leptini uygulanan gruplar olusturdu. Leptin hormonu 6 gun sureyle deri alti olarak uygulandi. Deney suresince tum gruplardaki farelerin bireysel canli agirliklari ile yem-su tuketimlerinin nasil degistigi belirlendi. Deney sonunda her farenin kalbinden 500 µl EDTA ile pihtilasmasi engellenmis kan alindi. Alinan kanlardan leptin analizi ve lenfosit alt tipleri analizleri yapildi. Elde edilen verilerden lenfosit alt tiplerinin miktarlari ve leptin seviyeleri tek yonlu varyans analizi ile degerlendirildi. Alti gunluk leptin uygulamasi doneminde vucut agirlik degisimleri tekrarlayan olcumlerde iki yonlu varyans analizi ile degerlendirildi.Deneyin adaptasyon doneminde farelerde hafif bir kilo artisi belirlendi. Alti gunluk leptin uygulamasi doneminde L100, L250 ve L500 gruplarinda birinci ve/veya ikinci gunden itibaren, L1000 grubunda ise leptin uygulamasinin hemen baslangicindan itibaren hafif bir vucut agirlik azalmasi belirlendi. Bu agirlik azalmasinin L1000 grubunda dorduncu gune kadar oldugu ve bu gruptaki vucut agirliklarinin tekrar eski seviyelerine ulastigi belirlendi. Ayni donemde leptinin yem tuketimlerini 2. gunden itibaren su tuketimlerini ise 1. gunden itibaren hafif olarak azalttigi bulundu. En yuksek leptin doz grubu olan L1000 grubunda CD4+ hucrelerin yuksek (p 0,05), yuzde olarak dogal oldurucu hucreler (% NK), CD8+ ve CD19+ hucrelerin ise leptin uygulamasindan etkilenmedigi belirlendi. AbstractThe aim of this study was to investigate effects of leptin, mainly produced by the adipose tissue and has pleiotropic effects, on food/water intake and lymphocyte subpopulations count in mice. Initially fourty healthly male Swiss albino mice were randomly divided into five groups. Mice in group I (group KFTS, control) were given serum physiologic and group II (group L100), III (group L250), IV (group L500), and V (group L1000) were given 100, 250, 500 ve 1000 µg/kg/day recombinant mouse leptin, respectively. Leptin were injected by subcutan for the next 6 days in groups II, III, IV, and V. Mice were monitored and weighed everyday during the experimental period. Daily food-water intake was recorded for each group. At the end of the study, blood samples (500 µl) with EDTA (as an anticoagulant) were obtained via intracardiac punction. Leptin levels and lymphocyte subpopulations in blood samples were analyzed. Lymphocyte subpopulations and leptin levels were examined by one-way analysis of variance (ANOVA). Changes of body weight and food-water intake during leptin administration period were evaluated with two-way ANOVA for repeated measures.Mean body weight and food intake slightly increased during the adaptation period. During the 6 days leptin administration period the mean body weight slightly decreased starting from the first and/or second day of the experiment in group L100, L250, and L500. In addition, the mean body weight in group L1000 also decreased from the starting day to day 4 of leptin administration. At the same period, food (from the second day of leptin administration) and water (from the first day of leptin administration) intake also decreased slightly. CD4+ cells markedly (p 0,05). Among all groups, no difference was found in the percentage of natural killer cells (NK%) and the amount of CD19+ and CD8+ cells." @default.
