- W2155491229 abstract "Thank you for giving me the opportunity to respond to Dr. Brodie et al.'s letter [ [1] Brodie M.J. et al. Letter to the editor: Antiepileptic drug therapy: does mechanism of action matter?. Antiepileptic drug therapy: does mechanism of action matter?. Epilepsy & Behav. 2011; 21: 342-343 Abstract Full Text Full Text PDF PubMed Scopus (16) Google Scholar ]. The reason I wrote the commentary was to add a perspective that I thought was missing in their article [ [3] Schmidt D. Commentary to Brodie et al. Antiepileptic drug therapy: does mechanism of action matter?. Epilepsy & Behav. 2011; 21: 342-343 Abstract Full Text Full Text PDF PubMed Scopus (16) Google Scholar ]. The authors, some of whom were instrumental in the development of antiepileptic drugs in the last 30 years or so, concentrated on conventional preclinical AED development. However, while conventional preclinical AED development revolves around finding new, or more often refined, mechanisms of drug action that influence seizure-generation, the sad reality is that the third and fourth generation fruit of AED development has failed to deliver substantially more efficacious drugs for common epilepsies compared to major second generation AEDs such as carbamazepine and valproate [ [4] Löscher W. Schmidt D. Modern antiepileptic drug development has failed to deliver: ways out of the current dilemma. Epilepsia. 2011; 52: 657-678 Crossref PubMed Scopus (388) Google Scholar ]." @default.
