- W2162566273 abstract "Amplified Fragment Length Polymorphism (AFLP) was used to study genetic relationship among leaf rust fungi (Melampsora spp.) occurring on Salix species in short-rotation coppice. Special interest was paid to a newly identified rust fungus found on S. viminalis in southern Sweden, morphologically similar to M. larici-epitea but with a distinct DNA profile. Genetic distances among 40 Melampsora isolates collected from S. viminalis, S. dasyclados, S. viminalis x dasyclados, S. daphnoides, S. acutifolia and M. larici-populina were calculated based on 101 AFLP markers. Neighbour-joining analysis revealed the presence of six clusters, which corresponded exactly to predefined groups, namely three formae speciales of M. larici-epitea, a stem-infecting form of Melampsora on S. viminalis, the newly identified Melampsora on S. viminalis and a group consisting of two isolates from the poplar rust M. larici-populina. All six clusters were well supported by bootstrap analysis (84 to 100% support). The newly identified Melampsora on S. viminalis was indicated to be genetically separated from M. larici-epitea as well as from the stem-infecting form of Melampsora. La technique Amplified Fragment Length Polymorphism (AFLP) a été utilisée pour étudier les relations génétiques chez les rouilles foliaires (Melampsora spp.) des saules qui sévissent dans les taillis à courte rotation. Une attention particulière a été portée à la rouille récemment identifiée sur Salix viminalis dans le sud de la Suède, qui est morphologiquement semblable àMelampsora larici-epitea mais différente pour le profil de l'ADN. Les distances génétiques chez 40 isolats de Melampsora récoltés sur S. viminalis, S. dasyclados, S. viminalis x dasyclados, S. daphnoides, S. acutifolia, et M. larici-populina, ont été calculées pour 101 marqueurs AFLP. L'analyse par ‘Neighbor-joining’à révélé l'existence de six groupes qui correspondaient exactement aux groupes prédéfinis, c'est-à-dire trois formes spéciales de M. larici-epitea, une forme de Melampsora infectant les tiges de S. viminalis, le nouveau Melampsora de S. viminalis, et un groupe composé des deux isolats de la rouille du peuplier M. larici-populina. Le nouveau Melampsora de S. viminalis s'est montré génétiquement distinct de M. larici-epitea et de la forme caulinaire de Melampsora. Die genetische Beziehung zwischen Blattrostpilzen (Melampsora spp.), die auf Salix -Arten in Energieplantagen vorkommen, wurde mit Hilfe von Amplified Fragment Length Polymophism (AFLP) untersucht. Besondere Aufmerksamkeit wurde einem neu identifizierten Rostpilz geschenkt, der auf Salix viminalis in Südschweden gefunden wurde und der morphologisch M. larici-epitea gleicht, aber ein anderes DNA-Profil aufweist. Basierend auf 101 AFLP Markern wurden die genetischen Distanzen bei 40 Melampsora-Isolaten berechnet, die von folgenden Salix-Arten gesammelt worden waren: S. viminalis, S. dasyclados, S. viminalis x dasyclados, S. daphnoides und S. acutifolia. Ausserdem wurde Melampsora larici-populina in die Untersuchung einbezogen. Die Cluster-Analyse ergab sechs Gruppen, die exakt mit den zuvor definierten Gruppen korrespondierten, nämlich drei formae speciales von M. larici-epitea, einer den Stamm infizierenden Form von Melampsora auf S. viminalis, der neu identifizierten Melampsora auf S. viminalis und einer Gruppe, die aus zwei Isolaten des Pappelrostes M. larici-populina bestand. Alle sechs Cluster wurden durch Bootstrapping gut bestätigt (84–100% Support). Die neu identifizierte Melampsora auf S. viminalis war gentisch von M. larici-epitea ebenso getrennt wie von der stamminfizierenden Form von Melampsora." @default.
