Thisprobe was specific forT.vaginalis DNA.Itrecognized strains fromtworegions inItaly (Sardinia, Piemonte) andfromMozambique (Africa). Inaddition, ourprobe didnotcross-react with bacterial (Escherichia coli, Enterococcus spp.,groupB streptococci, Gardnerella vaginalis, Neisseria gonorrhoeae, Chiamydia trachomatis, andLactobacillus spp.), viral (herpes simplex virus type2),fungal (Candida albicans), protozoan (Entamoeba histolytica, Giardia lamblia, Plasmodium falciparum, Leishmania major, and Leishmania infantum), or humannucleic acids. Theprobereacted withPentatrichomonas hominis and Trichomonas foetus. Thelimit signal recognized byour probecorresponded totheDNA of200T.vaginalis isolates. The2.3-kb probe wasusedinaclinical analysis of98samples. Ofthese, 20samples were foundtobe positive bothwiththeprobe andbycell culture, andonly14ofthese were positive bya standard wetmount method. Trichomonas vaginalis isresponsible forhumantrichomoniasis (for areview, seereference 11), oneofthemost widespread sexually transmitted diseases intheworld, with anincidence of10to50%insexually transmitted disease clinics (26). Accurate diagnosis isnecessary forspecific treatment andforthecontrol oftheinfection. Onemethod of diagnosis isidentification ofthemotile flagellate protozoan inavaginal discharge bydirect microscope examination (wet mount). Bythis method itispossible toidentify T.vaginalis in50to70%ofinfected patients (26). Several methods have beendeveloped forimproving detection ofT.vaginalis in clinical samples.
