- W2171950295 abstract "The liver as a central metabolic organ and part of the body’s own immune defence is inextricably linked with the acute phase reaction (APR) of a systemic immune reaction. Especially with respect to the breakdown of toxic substances resulting from tissue damage, liver cells play a key role. The melanocortin-4-receptor (MC4R) as a mediator of anti-inflammatory signals acts as an inhibiting factor against the production of inflammatory cytokines. The objective of this research project was both an examination of the distribution of the MC4R during the APR in the liver, as well as an evaluation of its pathophysiological significance therein. In a rat model, an APR was induced by intramuscular injection of turpentine oil (TO). At defined points of time after the injection, RNA was extracted from both liver and brain tissues and serum samples. In addition, immunostaining of tissue sections was performed and proteins were isolated. The presence of the MC4R in the rat liver could be established using PCR and Western Blot analyses as well as immunofluorescence and peroxidase stainings. In comparison with an untreated cohort of control animals, a significantly raised protein expression of the MC4R could be shown at 4 hours, 6 hours and 12 hours after APR induction. Immunohistology established the presence of the MC4R in the nucleus of parenchymal and non-parenchymal liver cells. Furthermore an expression shift from the cytoplasm to the nucleus was observed. MC4R was also shown to be present in Kupffer cell and hepatocyte cultures. In summary, the present work demonstrates that the MC4R is not only expressed in the brain but also in the liver of rats. Its expression is increased after APR induction. We assume that MC4R-mediated signal transduction pathways play a significant regulatory role by limiting the synthesis of pro-inflammatory cytokines during the APR." @default.
