In 2011, production was up to 170,000 tons from 8,000 ha of planting in Mengzi, Yunnan. During our survey of pathogens of pomegranate in Mengzi from autumn 2011 to autumn 2013, Chaetomium globosum Kunze was isolated frequently from leaf spots collected in June to September of each year. Leaf spots appeared as scattered, small, circular, purple or reddish-brown spots. Each spot gradually increased in size and turned dark red or black at the center while remaining yellow on the edges. Infected leaves surrounding the spots turned yellowish or yellow and an average of 30% of these leaves dropped. A pure culture (Bd-2) was obtained by tissue isolation and single-spored on potato dextrose agar (PDA) at 25°C. The colony on PDA medium was up to 90 mm in diameter after 6 days at 25°C. Initially, the colony was white and turned greenish with age. The ascomata were covered with helical appendages, with a papillate ostiole, globose or obovoid, green, 180 to 240 µm in diameter, and 300 to 440 µm high (n = 50). The conidia were greenish and lemon-shaped, 7 to 9 × 8 to 12.5 µm (n = 50). These morphological characteristics were consistent with the morphology of C. globosum (Lan et al. 2011). A spore suspension (1 × 106 conidia/ml) was prepared by harvesting conidia from two-week-old cultures grown in the dark at 25°C, then a 30-µl drop of spore suspension was placed on three healthy leaves of one 2-year-old seedling nonwounded or wounded with a sterilized needle. Thirty µl of sterile water was placed on three healthy leaves on another seedling and used as the control. Each treatment had five replicates. All seedlings were covered with plastic bags and placed outdoors at 12 to 26°C. Similar symptoms to those observed in the orchards developed on all the wounded leaves inoculated with Bd-2 after seven days and on 66.7% of the nonwounded leaves two to three weeks after inoculation, but not on the control leaves. The pure culture reisolated from the infected leaves was identical to the original colonies and conidia, completing Koch’s postulates. The internal transcribed spacer (ITS) sequence of rDNA of the pathogenic strain Bd-2 (GenBank Accession No. KJ183693) had 100% identity with C. globosum strain SL-9 (KC460815) and isolate 2987 (FJ224101). Molecular and morphological observations all showed that the strain Bd-2 was C. globosum. To our knowledge, this is the first report of C. globosum on pomegranate from Yunnan, China as well as worldwide. A leaf spot disease on potato (Seth 1970) and ramie (Boehmeriae nivea) (Zhang and Zhang, 1988) caused by C. globosum has been reported previously.References:Lan, N. 2011. J. Huazhong Agric.
