- W2181273178 abstract "Recent advances in the field of exocytotic secretion have depended almost exclusively on the use of permeabilized cells that allow the composition of the cytosol to be precisely controlled. Many agents and strategies for plasma membrane permeabilization have been utilized (see Table 1), and it is impor tant to appreciate that these generate, in the plasma membrane of individual cell types, lesions having widely differing dimensions and lifetimes. For example, the exocytotic release of catecholamines has been investigated using bovine adrenal chromaffin cells permeabilized by high voltage electric dis charge (HVD) (37), by treatment with plant glycosides (19, 60), staphylococ cal a-toxin (l, 4) and by streptolysin-O (SL-O) (64), and by capacitance measurement (to monitor the increase in plasma membrane surface area) using a patch-pipette in the whole cell mode (49). Two of these permeabiliz ing procedures (HVD and a-toxin) generate small membrane lesions allowing the dialysis but not full wash-out of the cytosol, while treatment with digito nin and SL-O (and presumably the patch-pipette) permit the release of cytosol proteins, though they do so at very different rates (60).
