- W2182100079 abstract "Heart and Skeletal muscle inflammation (HSMI) was first recorded in farmed Atlantic salmon (Salmo salar) in Norway in 1999. Since 1999 there have been an increasing number of outbreaks and the problem seems to be spreading along the Norwegian cost. Currently the disease is considered to have great economical impact for the Norwegian salmon farming industry. Outbreaks have been registered throughout the year, but most frequently occurring 5 to 9 months after seawater transfer. The mortality has varied from slightly enhanced to 20%. The most consistent findings are severe and extensive inflammation in heart and skeletal muscle. Kongtorp et.al. (2004) showed that the disease could be induced by injecting homogenized tissue from fish diagnosed with HSMI, and that the disease was transferred by cohabiting challenge. The diagnosis was verified by histological examination, but the ethological agent has not yet been cultured in vitro. This is the first report of propagated virus in cell culture. Tissue was collected from a clinical outbreak of HSMI and homogenized prior to clarification and 0.45 µm filtration. Clarified homogenate was inoculated onto cell cultures. Supernatant and cell lysate was collected 14 days p.i. and inoculated onto fresh cell cultures. Sensitivity to chloroform was determined by adding chloroform to the virus suspension to a final concentration of 10% (v/v). The mixture was shaken for 30 minutes at RT and centrifuged to remove the chloroform prior inoculation of cell cultures. The cell cultures were kept for 14 days before examination for cytopathic effect. Atlantic salmon (50 g) were injected with tissue homogenate from HSMI diagnosed fish, virus derived from cell culture or PBS. The fish was kept in seawater at 12°C, and organ tissue were collected 8 weeks p.i. Heart tissue was submerged in 10% phosphate buffered formalin for histology or 2% glutaraldehyd for transmission electron microscopy." @default.
